Construction of the Physical Map of Homo sapiens -------------------------------------------------------------------- The physical map of Homo sapiens was constructed by generating fingerprints from several BAC libraries, but mostly RPCI-11 and RPCI-13 (http://bacpac.chori.org), and CalTech B, C, and D (http://informa.bio.caltech.edu/idx_www_tree.html) libraries. The map consists of 415,801 BAC clones with an average insert size of 172kb. The clones were digested and run on agarose gels to produce fingerprints, analyzed with Image (http://www.sanger.ac.uk/Software/Image), and assembled in an FPC database (http://www.agcol.arizona.edu/software/fpc/). The assembled contigs were manually edited to achieve proper order based upon fingerprint banding patterns and were then merged with one another to generate the largest contigs possible. Contigs within the map database have been numbered with respect to their position along the chromosome beginning with the first contig in the cytogenetic band. The first digit (or two digits) represents the chromosome that the contig is anchored to. A '9' in the second digit (or third digit) location indicates an unanchored contig on that chromosome. For example, chromosome 1 contigs are numbered from 1001-1048 and the unanchored contigs on chromosome 1 are from 1949-1959. Chromosome X contigs start with 23, chromosome Y with 24 and unlocalized contigs with 25. The physical map currently consists of 528 contigs. This physical map is a work-in-progress and as such is subject to change as edits are needed. Occasional updates of the database will be made available. The conversion of the Fpc sequence tags to WUGSC sequnce status is as follows. This is not useful to most users outside WUGSC.
