Neuroinflammation driven by reactive glia and immune cell infiltration is a hallmark of many neurodegenerative diseases and brain aging. However, there remain gaps in our knowledge on the causal mechanisms by which reactive glia and peripheral immune cells modulate neuronal function in aging and disease. Recently developed in vivo and iPSC based CRISPR KO screening approaches reveal genetic regulators of microglial-neuron cross talk and immune cell infiltration, providing mechanistic insight and potential therapeutic targets for neurodegenerative diseases and aging.
